(2S)-3-amido-propanyl-amino-bis(m-PEG8)-2-maleimido-acid-(Val-Cit-PAB-MMAE) is a novel branched Maleimide-VC-PAB-MMAE linker-payload molecule designed to address several common shortcomings of conventional ADC designs, including poor physicochemical properties and linker-driven DAR instability. The maleimide moiety reacts with thiol groups on the antibody, while two PEG8 chains not only helps reduce aggregation, but also accelerate maleimide ring-opening hydrolysis to increase the conjugate stability. In addition, the PEG chains increase aqueous solubility.
