Ibrutinib
Ibrutinib (PCI-32765) is a selective, irreversible BTK inhibitor (IC50 = 0.5 nM) that blocks B cell activation, induces apoptosis, and prevents tumor microenvironment homing. It inhibits BTK autophosphorylation (IC50 = 11 nM) and downstream signaling, with clinical applications in B cell malignancies like mantle cell lymphoma and chronic lymphocytic leukemia.
