IR-117-17 is an ionizable cationic and biodegradable amino lipid used in the formulation of lipid nanoparticles (LNPs). Nebulized LNPs containing IR-117-17, the cationic lipid 1,2-dioleoyl-2-trimethylammoniumpropane (1,2-DOTAP), and other components, encapsulating reporter mRNA, induce luciferase luminescence in the lungs and nasal passages of mice. Similarly, nebulized LNPs encapsulating Cre mRNA trigger Cre expression in cells of the large and small airways, alveolar macrophages, and epithelial cells in Ai14 mice, which are genetically modified to express the fluorescent protein tdTomato upon Cre-mediated translation.
