L-threo-sphingosine (d18:1), an analog of D-erythro-sphingosine, inhibits protein kinase C with 50% inhibition at 2.2 mol%, making it slightly more potent than D-erythro-sphingosine (50% inhibition at 2.8 mol%) and other analogs with shorter alkyl chains. Unlike D-erythro-sphingosine, L-threo-Sphingosine C-18 does not reduce serine palmitoyltransferase (SPT) activity, the rate-limiting enzyme in ceramide biosynthesis, in primary cultured cerebellar cells.
