Mal-amido-PEG8-val-gly-PAB-PNP is an ADC linker comprised of a terminal maleimide, PEG chain for enhanced water-solubility, Val-Gly dipeptide, PAB group, and PNP group which can be substituted by amine-bearing molecules. The maleimide is applied to covalently bind free thiols on the cysteine residues of proteins. The Val-Gly dipeptide is cleaved by cathepsin enzymes within the cell to allow for efficient payload delivery.
