Ionizable lipids are a class of lipid molecules that remain neutral at physiological pH but become protonated - and thus positively charged - under acidic conditions. These lipids are a key component of lipid nanoparticles (LNPs), which are widely used for the systemic delivery of RNA therapeutics. Their pH-sensitivity is particularly advantageous for mRNA delivery in vivo, as neutral lipids exhibit minimal interaction with the anionic membranes of blood cells, thereby enhancing the biocompatibility of LNPs.
