Overview
MagicBrite ECL DuraPlus Substrate Kit is a sensitive, enhanced luminol-based chemiluminescent substrate (ECL) for the detection of horseradish peroxidase (HRP) in immunoblotting and other immunoassay applications. The ECL substrate detects high femo gram amounts of antigen with stable and long light output (5 to 24 hours), which can be visualized using X-ray films or CCD-based imagers. Appropriate primary and secondary antibody dilutions are suggested for attaining optimal signal intensity and duration.

The indicated amount of BSA( 50ng to 2ng) was immobilized on a nitrocellulose membrane and incubated with MagicBrite DuraPlus ECL Substrate (A) and SuperSignal extended Dura(B):
anti-BSA goat IgG: 1:2000; rabbit anti-goat HRP antibody : 1:80K, ChemiDoc XRS+, 4 seconds exposure. All experiments were performed in duplicate.
Signal duration comparison by slot-blot: Rabbit anti-goat-HRP antibody (catalog # BP-50081) was immobilized on a nitrocellulose membrane and detected with MagicBrite ECL DuraPlus(A) and SuperSignal extended Dura(B) with an imaging system (ChemiDoc XRS+, 180 seconds exposure).
