Mc-Phe-Lys(Boc)-PAB is a linker featuring a maleimide, a Phe-Lys dipeptide with a Boc-protected ε-amine, and a PAB group on its N-terminal. Maleimide is a thiol-specific covalent group which is used to label reduced cysteines in proteins, while the benzylic alcohol on the PAB can be ligated to payloads such as MMAE or MMAF. Finally, the N-Boc protecting group can be removed to allow for reactivity at the lysine’s ε-amine.
